Other than general debate week, things are pretty much as normal. This year the general debate is the week starting Mon Sept 22 and that is when there will be street closings. The two absolute WORST days of that week will be the Tues and Weds as those are the days the U S president and other top world leaders give their speeches so security will be the tightest as they move around town. Security will also be tight and disruptive around any hotel where a top world leader is staying and any location where they are attending a social or other event (many of these and they can be just about anywhere in
Manhattan). In recent years the US president has stayed at the Barclay at 48th and Lex. The Fri and Sat things will be getting back to normal. I live in the security zone and generally count on about a week of street closings, traffic jams and inconvenience (e.g., having to have ID handy to get home).
I would say greatest likelihood of disruption/delay is anywhere from 34th to 57th streets from 6th Av to East River, but can be anywhere there is a UN related event, including area around Met Museum if and when there is event there. You should check with UN to find out the period when tours are suspended.
